After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture uptake. Residual water content is measured because small changes can alter chemical stability and cake appearance. Storage temperature depends on the material; many biological products are kept at 2–8 °C, while some require −20 °C or colder. Exposure to ambient humidity during handling can cause the porous solid to absorb water and collapse. Container closures and stoppers are therefore selected for low moisture transmission and compatibility.
Quality control for lyophilized materials includes visual inspection of the cake, measurement of residual moisture, and tests for reconstitution time. An acceptable cake is typically uniform and may be slightly porous; shrinkage, meltback, or cracks can indicate process deviations. Analytical methods such as Karl Fischer titration, thermogravimetric analysis, and near-infrared spectroscopy quantify water content. Reconstitution time is recorded because a very slow or incomplete dissolution can signal collapse or aggregation. Stability studies compare samples stored under defined temperature and humidity conditions over months or years.
| Property | Value | Notes |
|---|---|---|
| Appearance | Porous cake or plug | Uniform structure suggests the drying cycle preserved the matrix. |
| Reconstitution time | Usually under 2 minutes | Depends on cake porosity, diluent volume, and excipient composition. |
| Water content range | 0.5–3% w/w | Common specification range; exact limits are product-specific. |
| Headspace oxygen | <1% v/v | Inert gas backfill reduces oxidation of sensitive materials. |
| Storage temperature | 2–8 °C or controlled room temperature | Choice depends on accelerated and real-time stability results. |
Quality control for lyophilized materials examines appearance, reconstitution time, residual moisture, and mechanical integrity. An acceptable cake is usually uniform and porous, though appearance alone does not prove stability. Karl Fischer titration is a common method for water content, while differential scanning calorimetry can reveal glass transition events. Stability studies track potency, aggregation, and moisture over time under defined temperature and humidity conditions. Specifications are product-specific and may include sterility and endotoxin tests for sterile preparations.
Misconceptions about lyophilization include the idea that dried products are indefinitely stable or that vacuum sealing eliminates all degradation. Chemical reactions can continue in the solid state, and some proteins lose activity even at low moisture. Another misconception is that any freeze-dryer cycle can be scaled by time alone; heat and mass transfer differ with equipment and load. Open questions remain about predicting long-term stability from short accelerated studies, particularly for amorphous formulations. Real-time stability data remain the standard for shelf-life assignment.
Stability programs monitor lyophilized products under defined temperature and humidity conditions over time. Real-time studies at recommended storage conditions are the reference, while accelerated studies provide early signals of degradation pathways. Because a dry cake can still undergo oxidation, hydrolysis, or aggregation, stability depends on residual moisture, excipients, and container headspace. Open questions include how best to predict long-term stability from short accelerated runs and how vial-to-vial variability affects shelf life. Current guidance treats these predictions as product-specific rather than universally generalizable.
Freeze-dried materials are hygroscopic to varying degrees and can take up moisture after drying. Storage therefore often uses sealed glass vials, rubber stoppers, and crimp seals to limit contact with ambient humidity. A desiccant may be included for moisture-sensitive products, although it is not universal. Controlled room temperature is sufficient for many lyophilizates, while others require refrigeration or freezing. Moisture ingress remains a primary cause of cake collapse, chemical degradation, and loss of reconstitution performance.
Quality assessment of a lyophilized product includes cake appearance, residual moisture, reconstitution time, and container closure integrity. A uniform, porous cake suggests that freezing and drying stayed within the formulation's design space. Cracks, shrinkage, meltback, or a glassy film can indicate thermal abuse or a formulation problem. Analysts also test for subvisible particles and sterility when the product requires those specifications. Visual inspection alone cannot confirm biological activity or chemical stability, so it is combined with analytical methods.
Quality control for freeze-dried lots combines visual inspection with instrumental tests. Cake appearance, color, and shrinkage are recorded against a reference, while residual moisture is measured by Karl Fischer titration or loss on drying. Thermal analysis can reveal phase transitions and crystallization events, and X-ray diffraction distinguishes amorphous from crystalline solids. Microbiological tests and container closure integrity checks are also routine for sterile products. Analytical methods must be validated for the matrix, because excipients and low moisture can affect accuracy. Open questions include how best to predict long-term stability from short-term data.
Lyophilized solids are often hygroscopic, so handling occurs in controlled low-humidity areas or glove boxes when the material is exposed. Vials remain sealed with elastomeric stoppers and aluminum crimps until use, because airborne moisture can raise residual water and shorten shelf life. The porous cake is fragile and may crack, shrink, or powder during transport. Personnel typically avoid repeated warming and cooling of sealed units, which can draw moisture through closures. These practices aim to preserve the low water content achieved during drying.
One barn is 10−28 square metres, about the cross-sectional area of a uranium nucleus. The name probably derives from early neutron-deflection experiments, when the uranium nucleus was described, and the phrases "big as a barn" and "hit a barn door" were used. Barn are typically used for cross sections in nuclear and particle physics. Additional units include the microbarn (or "outhouse") and the yoctobarn (or "shed").

These are various dosage forms and technologies which include but not limited to nanoparticles, liposomes, microneedles, and hydrogels that can be used to enhance therapeutic efficacy and safety. The same route can accommodate multiple dosage forms; for example, the oral route may involve tablet, capsule, or liquid suspension. While the transdermal route may use a patch, gel, or cream. Drug delivery incorporates both of these concepts while encompassing a broader scope, including the design and engineering of systems that operate within or across these routes. Common routes of administration include oral, parenteral (injected), sublingual, topical, transdermal, nasal, ocular, rectal, and vaginal. However, modern drug delivery continue to expand the possibilities of these routes through novel and hybrid approaches. Since the approval of the first controlled-release formulation in the 1950s, research into new delivery systems has been progressing, as opposed to new drug development which has been declining. Several factors may be contributing to this shift in focus. One of the driving factors is the high cost of developing new drugs. A 2013 review found the cost of developing a delivery system was only 10% of the cost of developing a new pharmaceutical. A more recent study found the median cost of bringing a new drug to market was $985 million in 2020, but did not look at the cost of developing drug delivery systems.

== History == The Tetra Brik package was introduced in 1963, after a long process of development and built on the previous Tetra Classic tetrahedron package that laid the foundation for Tetra Pak. Despite the revolutionary character of the new retail system, by the end of the 1950s Ruben Rausing and the Tetra Pak management team realised that the Tetra Pak package portfolio needed to be supplemented by an additional rectangular model to continue to be competitive. Large amounts were spent on development, and in 1963 the first Tetra Brik packaging machine was installed in Motala in central Sweden. Due to its effective use of space and materials and the increased efficiency in distribution and storage that resulted from the rectangular shape, the Tetra Brik soon became Tetra Pak's best seller in Sweden and internationally and paved the way for Tetra Pak's enormous success during the 1970s and onwards. The Tetra Brik package family has since been extended and is currently composed of Tetra Brik and Tetra Brik Aseptic in the shapes Base, Square and Edge, ranging from volumes of 80 to 2000 ml and with a number of different caps. Tetra Brik has been distinguished as one of the 20th century's design icons. It featured in the 2004 Museum of Modern Art New York exhibition Humble Masterpieces and in the 2011 London Science Museum/Vitra Design Museum exhibition Hidden Heroes – The Genius of Everyday Things. It is now part of the permanent collection at the MoMA in New York.

More recently, several groups have reported the crystal structure of Nampt/PBEF/visfatin and they all show that this protein is a dimeric type II phosphoribosyltransferase enzyme involved in NAD biosynthesis. eNAMPT has been shown to be more enzymatically active than iNAMPT, supporting the proposal that eNAMPT from adipose tissue enhances NAD+ in tissues with low levels of iNAMPT, notably pancreatic beta cells and brain neurons.
==== By mouth ==== The pain medication paracetamol (acetaminophen) or NSAIDs are first line treatments for osteoarthritis related pain. With paracetamol, pain relief does not differ according to dosage. However, a 2015 review found acetaminophen to have only a small short-term benefit with some concerns on abnormal results for liver function test. For mild to moderate symptoms effectiveness of acetaminophen is similar to non-steroidal anti-inflammatory drugs (NSAIDs) such as naproxen, though for more severe symptoms, NSAIDs may be more effective. NSAIDs are associated with greater side effects such as gastrointestinal bleeding. Another class of NSAIDs, COX-2 selective inhibitors (such as celecoxib) are equally effective when compared to nonselective NSAIDs, and have lower rates of adverse gastrointestinal effects, but higher rates of cardiovascular disease such as myocardial infarction. They are also more expensive than non-specific NSAIDs. Benefits and risks vary in individuals and need consideration when making treatment decisions, and further unbiased research comparing NSAIDS and COX-2 selective inhibitors is needed. The COX-2 selective inhibitor rofecoxib was removed from the market in 2004, as cardiovascular events were associated with long term use. Education is helpful in self-management of arthritis, and can provide coping methods leading to about 20% more pain relief when compared to NSAIDs alone. Failure to achieve the desired pain relief in osteoarthritis after two weeks of therapy should trigger reassessment of dosage and pain medication.
In some parts of Europe and India, Datura has been a popular poison for suicide and murder. From 1950 to 1965, the State Chemical Laboratories in Agra, India, investigated 2,778 deaths caused by ingesting Datura. A group called Thugs (practicers of thuggee) were reportedly devotees of an Indian religious cult made up of robbers and assassins who strangled or poisoned their victims in rituals devoted to the Hindu goddess Kali. They were alleged to employ Datura in many such poisonings, using it also to induce drowsiness or stupefaction, making strangulation easier. Datura toxins may be ingested accidentally by consumption of honey produced by several wasp species, including Brachygastra lecheguana, during the Datura blooming season. These semi-domesticated honey wasps apparently collect Datura nectar for honey production, which can lead to poisoning. The U.S. Centers for Disease Control and Prevention reported accidental poisoning resulting in hospitalization for a family of six who inadvertently ingested Datura used as an ingredient in stew. In some places around the world, such as India due to the Drug & Cosmetic Act 1940 & Rule 1995, buying, selling, or cultivating Datura plants is prohibited. Sealed vials or containers should be kept at the temperature specified by stability data, often controlled room temperature or 2–8 °C. Moisture and oxygen barriers are important because both can degrade sensitive materials. Opened containers may need immediate use or protection from ambient humidity.
It usually appears as a uniform porous plug or cake that fills the container without excessive shrinkage. Color should match the specification, and there should be no meltback or visible foreign matter. Minor cracking may be acceptable if the product still meets moisture and potency limits.
Water content is a key stability parameter because excess water can promote hydrolysis, aggregation, or cake collapse. It also affects reconstitution and product weight. Each product has a target range, and methods such as Karl Fischer titration are used to verify it.
Most are held in sealed containers at controlled temperatures, often 2–8 °C, while some require frozen storage. Protection from moisture and light helps preserve the dry matrix. Exact conditions are set by the manufacturer or study protocol.
